
More Potent CAR-T Cells via Akadeum BACS™ Microbubbles Integration with the Cytiva Sepax™ C-Pro Cell Processing System
This application note presents data from a fully closed, negative selection T cell isolation workflow integrating Akadeum Microbubbles with the Cytiva Sepax™ C-Pro Cell Processing System.
Using leukopak material from three common healthy donors in a model CD19 CAR-T process, the Sepax-integrated Akadeum workflow was evaluated and compared directly to an industry-standard in-bag magnetic CD4/CD8 positive selection method.
The study demonstrates that the Sepax/Akadeum workflow achieves comparable CD3⁺ purity while delivering higher T cell recovery relative to magnetic selection. Downstream characterization following CAR transduction and expansion showed trends toward improved expansion, favorable CD4/CD8 balance, reduced exhaustion, and increased stem-cell–like memory phenotypes. Functional assessment in co-culture assays further demonstrated cytotoxic activity against CD19-expressing target cells.
Importantly, these results were obtained within a single, fully closed Sepax workflow, without the need for external magnets or open processing steps, highlighting the potential advantages of a magnet-free, in-bag isolation approach for scalable and manufacturing-ready CAR-T workflows.
